Salivary microRNA Signatures Associated with Autism Spectrum Disorder inBrazilian Adults

Abstract

Autism Spectrum Disorder (ASD) is a complex human neurodevelopmental condition with increasing prevalence, yet early diagnosis remains challenging, particularly in low- and middle-income countries such as Brazil, where molecular studies are scarce. This study investigated the salivary expression of four microRNAs (miR-23a-3p, miR-140-3p, miR-151a-3p, and miR-486-3p) in 45 Brazilian adults (24 with ASD and 21 neurotypical controls), using RT-qPCR and bioinformatic analyses of functional enrichment and gene target prediction. The results demonstrated significant overexpression of miR-23a-3p (FC=28.8-fold; p=0.0012), miR-140-3p (p=0.0352), and miR-486-3p (p=0.0352) in the ASD group, as well as co-expression among these miRNAs. miR-23a-3p showed the best diagnostic accuracy (AUC=0.827; specificity=100%), while miR-486-3p exhibited differential expression associated with the year of diagnosis (p=0.012). A very strong correlation was observed between miR-23a-3p and miR-151a-3p expression (ρ=0.973), along with co-regulation of gene targets across salivary gland, prefrontal cortex, and hypothalamic tissues. Enrichment analyses revealed association of gene targets with processes central to neurodevelopment, including forebrain and telencephalon development, cognition, social behavior, synaptic plasticity, and synaptic vesicle cycling. These findings provide novel evidence of salivary microRNA dysregulation in Brazilian adults with ASD, a demographic notably underrepresented in molecular autism research. Collectively, the evidence indicates that specific microRNAs involved in synaptic regulation are dysregulated in ASD and highlights the potential of saliva as a non-invasive and accessible biofluid for molecular screening in neurodevelopmental disorders.

Marcos Fernandes Morales, Dom Bosco Catholic University

Holds a Bachelor's degree in Biological Sciences from the Catholic University Dom Bosco (2014–2017) and a Master's degree in Biotechnology from the same institution (2018–2020). He was a scholarship recipient of the Institutional Program for Scientific Initiation Scholarships (PIBIC) for two consecutive years, developing projects with an emphasis on Molecular Genetics of Microorganisms. He conducted research in the field of Forensic Microbiology, with experience in metagenomics and bioinformatics for metadata analysis (QIIME 2), as well as an internship at the Forensic Microbiology Laboratory at Alabama State University (AL/USA). He is currently a Ph.D. candidate in Biotechnology at the Catholic University Dom Bosco (2022–present; CAPES scholarship holder), focusing on the investigation of salivary microRNAs as potential biomarkers for the early identification of Autism Spectrum Disorder (ASD). His research involves absolute and relative quantitative analyses by RT-qPCR, integration of bioinformatic metadata, and validation of molecular candidates (using R/RStudio and Cytoscape). He actively participates in the miRTEA project for validation studies in molecular prognosis and in SNCT workshops at state schools in Mato Grosso do Sul, aimed at scientific, technological, and informational outreach on ASD. His main areas of concentration include: Forensic Microbiology, Metagenomics, Transcriptomics, Molecular Biology, Genetics, and Bioinformatics-based clinical screening biomarkers.

Carina Elisei de Oliveira, Dom Bosco Catholic University

Holds a Bachelor's degree in Biological Sciences from the Federal University of Juiz de Fora (1999), a Master's degree in Veterinary Sciences from the Federal Rural University of Rio de Janeiro (2001), and a Ph.D. in Veterinary Sciences from the Federal Rural University of Rio de Janeiro (2005). He is currently a professor and coordinator of the PIBIC program at the Catholic University Dom Bosco, and a research professor at the Catholic University Dom Bosco. He has experience in the field of Animal Parasitology, working on the development of diagnostic techniques for the detection of microorganisms/infectious agents in domestic and wild animals, with an emphasis on Molecular Biology, mainly focusing on the following topics: Plasmodium (Novyella) juxtanucleare in Gallus gallus, parasitology, dogs, the Pantanal, and hemoparasites.

Manoella Tomasine Schmidt, Dom Bosco Catholic University

Undergraduate student in Nutrition, expected completion: 2027/A, and in Psychology, expected completion: 2030/A, at the Catholic University Dom Bosco (UCDB). She is currently a scholarship recipient of the Institutional Program for Scientific Initiation Scholarships (PIBIC/CNPq-FUNDECT), 2025/2026 cycle, effective from September 2025 to August 31, 2026. She is developing activities through the Be.micro Research Center and the miRTEA Project, dedicated to science and innovation in ASD care. She has experience gained through extracurricular internships and teaching assistantships in the areas of Food and Nutrition Unit Administration (AUAN), Clinical Nutrition, and as a teaching assistant in Anatomy and Physiology. Holds ISAK Level 1 International Certification (International Society for the Advancement of Kinanthropometry), qualified to perform standardized anthropometric assessments, including measurement of skinfolds, circumferences, bone diameters, body mass, and height, contributing to clinical practice and health research.

Graduated in Biomedicine from the Catholic University Dom Bosco (UCDB), I hold a Master's degree in Biotechnology with an emphasis on Microbiology and Molecular Biology from the same institution. I am currently enrolled in the Ph.D. program in Biotechnology, under the supervision of Prof. Dr. Alinne Pereira de Castro, also at the Catholic University Dom Bosco (UCDB). During my academic training, I was a CNPq scholarship recipient for three consecutive cycles in the Institutional Program for Scientific Initiation Scholarships (PIBIC). I worked on scientific projects in the areas of microbiology and biochemistry, investigating the impact of arbuscular mycorrhizal fungi on starch production in arrowroot. Additionally, I conducted research in biochemistry and entomology, focusing on the prospection of compounds with insecticidal activity from microorganisms and plants from the Cerrado and Pantanal biomes. I am currently a member of the BeMicro research group, supervised by Prof. Dr. Alinne Pereira de Castro, which develops predictive and early diagnostic tools for the risk of Autism Spectrum Disorder (ASD) based on salivary biomarkers. I participate in projects funded by Fundect, including "Salivary microbiome and identification of biomarker microorganisms for early diagnosis of ASD" (coordinated by Dr. Nathalia Cavichiolli de Oliveira).

Julia Cunha Gonzales, Dom Bosco Catholic University

Holds a Bachelor's degree in General Pharmacy from the Federal University of Mato Grosso do Sul – UFMS (2013). Master's degree in Sciences – Concentration Area in Biosciences Applied to Pharmacy, from the Faculty of Pharmaceutical Sciences of Ribeirão Preto/USP (2017). Completed a Ph.D. through the Graduate Program in Biosciences Applied to Pharmacy at the Faculty of Pharmaceutical Sciences of Ribeirão Preto/USP (2023). Has experience in the areas of Photodynamic Therapy applied to fungi, and Bacteriology, with an emphasis on molecular epidemiology, virulence, and genomics of enterobacteria. Currently, she is a Postdoctoral Researcher at the Catholic University Dom Bosco (UCDB), in Campo Grande/MS, where she researches salivary microRNAs as biomarkers for the early diagnosis of Autism Spectrum Disorder (ASD), combining knowledge in molecular biology and public health.

Currently pursuing a Bachelor's degree in Biomedicine at the Catholic University Dom Bosco (UCDB). Member of a Scientific Initiation project since 2022, focused on validating salivary miRNAs as potential biomarkers for Autism Spectrum Disorder (ASD). Member of the Be.micro research group, composed of researchers from various health fields at UCDB, dedicated to developing multidisciplinary studies aimed at a better understanding and advancement of knowledge about Autism Spectrum Disorder.

Alinne Pereira de Castro, Dom Bosco Catholic University

Holds a Bachelor's degree in Biological Sciences from the Catholic University of Brasília, a Master's degree in Genomic Sciences and Biotechnology, and a Ph.D. in Molecular Biology from the University of Brasília (UnB). Since 2014, she has been a professor/researcher at the Catholic University Dom Bosco (UCDB), accredited in the Graduate Program in Biotechnology, and serves as a Municipal Councilor for Science, Technology, and Innovation (CMCTI) in the state of Mato Grosso do Sul. She is the founder and creator of the company mirTEA Diagnósticos, coordinating several projects.
